Time-Resolved Fluorescence HPLC Detection Using Semiconductor Light Sources: Principles and Applications

Abstract

The simultaneous static and time-resolved fluorescence detection in high performance liquid chromatography (HPLC) systems using time-correlated single photon counting (TCSPC) and modulation techniques is a very recent topic in the area of analytical and physical chemistry. Whereas most commercial systems use simple intensity detection in fluorescence systems or fluorescence spectra with very low sensitivity we use real-time time-resolved fluorescence counting with on-the-fly data evaluation and display like a typical chromatogram. The static measurement is the sum of counts in the time interval. Both results are independent and characteristic for the fluorescent analytes. Additionally they are of equal sensitivity. In order to achieve ns resolution modern semiconductor light sources (nowadays available from 235 nm to MIR) are used. They are fast, small, low cost, low energy consuming, durable, and reliable (no maintenance necessary). Modern electronics allow a setup in typical HPLC component size in contrast to older complicated lamp or laser applications. For the measuring procedure no adjustment or calibration for the chromatogram is necessary. A rapid modification for new applications, such as new developments for legal regulations, is possible due to the flexible modular design. The application is patent pending: DE 102010016801 A1.
