    The aim of this work was to develop a robust and inexpensive method of CO-Oximetry. A new method which determines the hemoglobin concentrations in a special wavelength range is introduced. It has been shown that minor demands for wavelength stability and bandwidth of the spectrometer are required. Thereby inexpensive and very small spectrometers can be used. Samples with different derivative compositions have been analysed with a measuring system especially developed, which included the required fluidics, optics, electronics and data recording. For the computation of the concentrations the multivariate procedures CLS (classical least square) and PLS (partial least square regression) were used. For the wavelength calibration of the spectrometer a new method was developed which permits to calibrate spectrometers with broad bandwidth more precisely than with conventional methods. Hence, not only the measured values of the peaks, but almost all measured values of a spectral lamp measurement can be used for calibration. The spectrometers are calibrated regarding an only once measured model spectrum. The underlying idea is to calculate the wavelength shift by cross-correlating corresponding blocks of the model spectrum and the spectrum measured with the spectrometer under calibration.
